To investigate the effects of the nitric oxide (NO) donor 2-(4-morpholinyl)-8-phenyl-4H-1-benzopyran-4-one (NOC-18) on blood pressure, heart rate, blood flow and vascular smooth muscle, we measured the blood pressure, heart rate and blood flow of the rat hind limb using a micro-occlusion technique. The vascular smooth muscle of the hind limb was studied with reference to its NO-induced relaxant activity. NOC-18 induced a dose-dependent reduction in blood pressure, and increased the heart rate in anesthetized rats. The increase in the heart rate induced by NOC-18 was not blocked by propranolol, but was blocked by bilateral vagotomy. NOC-18 induced a dose-dependent increase in blood flow. The increase in blood flow induced by NOC-18 was blocked by pretreatment with bilateral vagotomy, guanethidine or propranolol. In isolated vascular smooth muscle of the rat hind limb, NOC-18 induced an endothelium-independent relaxation. Thus, NOC-18 induces dose-dependent increases in heart rate and blood flow, and is likely to be an effective cardiotonic agent.
